目的:探讨Foxp3和RORγt在肝泡型包虫病患者肝脏组织中的表达及意义。方法:将手术治疗的14位肝泡型包虫病患者的肝脏组织分为3组:病灶组织组(L组)14例,病灶旁组织组(P组)14例,正常肝组织组(N组)14例。应用实时荧光定量PCR(q RT-PCR)法检测各肝脏组织中Foxp3和RORγt m RNA的表达,分析其结果。结果:Foxp3 m RNA在L组中的表达明显高于P组(P=0.005)和N组(P=0.004),P组中的表达高于N组(P=0.246)。RORγt m RNA在L组中的表达高于P组和N组(L组vs P组,P=0.187);L组vs N组,P=0.044),在P组中的表达高于N组(P=0.027)。肝脏不同组织中,RORγt与Foxp3的表达量呈正相关。结论:Th17细胞转录因子Foxp3和Treg细胞转录因子RORγt在肝脏病灶组织中m RNA的表达升高,这些变化可能与泡型包虫病发病过程中寄生虫肉芽肿的形成有关。
Objective: To investigate the expression and significance of Foxp3 and RORγt in hepatic tissues of patients with hepatic hydatid disease. Methods: The hepatic tissues of 14 patients with hepatic echinococcosis were divided into three groups: 14 cases in the lesion group (L group), 14 cases in the para-focus group (P group), 14 cases in the normal liver tissue group (N Group) in 14 cases. The expression of Foxp3 and RORγt m RNA in each liver tissue was detected by real-time quantitative PCR (q RT-PCR), and the results were analyzed. Results: The expression of Foxp3 mRNA in group L was significantly higher than that in group P (P = 0.005) and group N (P = 0.004). The expression of Foxp3 mRNA in group P was higher than that in group N (P = 0.246). The expression of RORγt mRNA in L group was higher than that in P group and N group (L group vs P group, P = 0.187); L group vs N group (P = 0.044) P = 0.027). In different tissues of liver, the expression of RORγt and Foxp3 were positively correlated. CONCLUSION: The expression of m RNA in Th17 cell transcription factor Foxp3 and Treg cell transcription factor RORγt is increased in hepatic lesions. These changes may be related to the formation of parasite granuloma during the pathogenesis of the disease.
