目的:研究血管内皮细胞在致动脉粥样硬化因子作用下,差异表达基因的结构与功能。 方法:氧化低密度脂蛋白(OX-LDL)诱导培养的人脐静脉内皮细胞,用差异显示-多聚酶链式反应(DDRT-PCR)技术,克隆表达水平有明显差异的基因片段,并用Northen印迹法证实阳性结果。 结果:发现OX-LDL诱导培养的人脐静脉内皮细胞(HUVEC)出现一些上、下调节的基因片段。已知的上调基因包括人的胸腺素(Thymosin)β4、细胞间粘附因子(ICAM)-1、FK506结合蛋白、ERp72;下调的基因有细胞色素B561、Restin。用Northern印迹证实了DDRT-PCR的结果,不同的上、下调节基因表达水平变化在30%~200%之间不等。 结论:本研究首次用DDRT-PCR的方法证实在体外OX-LDL可诱导HUVEC许多基因表达水平的变化。细胞中细胞间粘附因子的高表达,进一步说明细胞间粘附因子是参与动脉粥样硬化过程重要的粘附分子。胸腺素β4基因的上调表达与OX-LDL诱导细胞肌动蛋白张力纤维的生成增加有关,可能影响内皮细胞的增殖。
OBJECTIVE: To study the structure and function of differentially expressed genes in vascular endothelial cells under the action of atherogenic factors. Methods: Human umbilical vein endothelial cells (HUVECs) were induced by oxidized low-density lipoprotein (OX-LDL). The differentially expressed genes were cloned by differential display-polymerase chain reaction (DDRT-PCR) Confirmed positive results. Results: Some up-and down-regulated gene fragments were found in OUV-induced human umbilical vein endothelial cells (HUVECs). Known up-regulated genes include human Thymosin β4, intercellular adhesion molecule (ICAM) -1, FK506-binding protein, ERp72; down-regulated genes are cytochrome B561, Restin. The results of DDRT-PCR were confirmed by Northern blotting. The expression levels of different up and down regulated genes ranged from 30% to 200%. Conclusion: This study is the first to confirm the changes of many gene expression in HUVEC induced by OX-LDL by DDRT-PCR. The high expression of intercellular adhesion molecules in cells further indicates that intercellular adhesion molecules are important adhesion molecules involved in the process of atherosclerosis. Upregulation of thymosin β4 gene expression and OX-LDL-induced increase in the production of actin fibers, may affect the proliferation of endothelial cells.
