目的:在明确解毒祛瘀滋阴药能够防治狼疮鼠类固醇性骨质疏松症的基础上,探讨其君药生地黄对狼疮鼠类固醇性骨质疏松症的疗效及可能的作用机制。方法:27只MRL/Ipr小鼠被随机分成空白组、模型组、生地黄组,每组9只。灌胃给药8周后,处死,取左上肢近端,运用小动物成像系统中的X线检测功能检测骨密度。取右上肢近端,制成病理切片,HE染色,显微镜下观察病理组织情况及骨形态发生蛋白-7(BMP-7)含量。结果:与空白组比较:模型组骨密度均值显著下降(P<0.05),骨小梁变细,排列不整齐、断裂,造模成功。BMP-7蛋白含量总体上升;与模型组比较:生地组骨密度显著提升(P<0.05),骨小梁量增多,排列较整齐。BMP-7蛋白含量总体上升。结论:解毒祛瘀滋阴药中君药生地对狼疮鼠类固醇性骨质疏松的疗效确切;其作用机制可能与其增加骨髓BMP-7的含量有关。
OBJECTIVE: To investigate the curative effect and possible mechanism of Herba Rehmanniae Decoction on steroid-induced osteoporosis of lupus in rats based on the clear detoxification and removing blood stasis nourishing drugs can prevent and cure steroid-induced osteoporosis in lupus mice. Methods: 27 MRL / Ipr mice were randomly divided into blank group, model group and radix rehmanniae group with 9 in each group. Eight weeks after gavage, they were sacrificed, the proximal left upper extremity was taken, and the bone mineral density (BMD) was measured by X-ray detection in a small animal imaging system. Take the proximal right upper extremity, made pathological sections, HE staining, under the microscope pathological conditions and bone morphogenetic protein -7 (BMP-7) content. Results: Compared with the blank group, the average BMD of the model group decreased significantly (P <0.05), and the trabecular bone became thinner, arranged irregularly, ruptured and the model was established successfully. Compared with the model group, the BMD of BMP-7 increased significantly (P <0.05), and the amount of trabecular bone increased. BMP-7 protein content increased overall. Conclusion: Jiedu Quyu Ziyin drug Jun medicine habit of lupus steroid osteoporosis exact effect; its mechanism may be related to its increased bone marrow content of BMP-7.
