目的研究成都地区献血者人群HCV感染的基因型分布。方法从血液筛查出的抗-HCV反应性血清标本中提取HCV RNA,RT-PCR获得HCV核酸C区及NS5B区序列片段,用MEGA软件构建序列进化树,分析HCV的亚型分布特征及可能的原因。结果对313例HCV RNA阳性标本分型结果如下,1b亚型283例,2a亚型14例,3b亚型7例,3a亚型3例,6a亚型5例,6u亚型1例;4和5基因型没有发现。结论在成都地区献血者人群感染的HCV中共有6个亚型分布。其中1b是绝对优势亚型,占90.41%(283/313);6u亚型在成都地区首次被检测到。
Objective To study the genotype distribution of HCV infection in blood donors in Chengdu. Methods The HCV RNA was extracted from the blood serum of anti-HCV reactive sera and the sequences of HCV nucleic acid C region and NS5B region were obtained by RT-PCR. The sequence evolutionary tree was constructed by MEGA software to analyze the subtypes of HCV and its possible distribution s reason. Results There were 283 cases of 1b subtype, 14 cases of 2a subtype, 7 cases of 3b subtype, 3 cases of 3a subtype, 5 cases of 6a subtype and 1 case of 6u subtype in 313 HCV RNA positive samples And 5 genotypes were not found. Conclusion There are 6 subtypes of HCV in the blood donors in Chengdu. Among them, 1b was the absolute predominant subtype, accounting for 90.41% (283/313); the 6u subtype was first detected in Chengdu.
