以苔酚蓝拒染法、核黄素-NBT还原法、DTNB还原法及RNA斑点杂交分析法分别对经不同浓度Isoverbascoside(Isov)处理的MGC80-3 细胞的生长速率、II期抗氧化酶(超氧化物歧化酶、谷胱甘肽过氧化物酶和过氧化氢酶)活性和N-ras、C-m yc及p53 基因表达作了检测.结果显示:经Isov处理后,细胞生长明显受抑;II期抗氧化酶活性较处理前明显上升;癌基因N-ras、C-m yc的表达较处理前下降,而抑癌基因p53 表达上升. 表明Isov 的抑癌作用与诱导II期抗氧化酶活性升高和癌基因表达改变有关
The growth rate, phase II antioxidant enzymes of MGC80-3 cells treated with different concentrations of Isoverbascoside (Isov) were determined by erythrose blue exclusion, riboflavin-NBT reduction, DTNB reduction, and RNA dot blot analysis, respectively. Superoxide dismutase, glutathione peroxidase, and catalase activity and N-ras, Cm yc, and p53 gene expression were detected. The results showed that cell growth was significantly inhibited by Isov treatment. The activity of phase II antioxidant enzymes increased significantly compared with that before treatment; the expression of oncogenes N-ras and Cm yc decreased compared with that before treatment, while the expression of tumor suppressor gene p53 increased. This indicated that the anti-cancer effect of Isov and the induction of phase II antioxidant enzyme activity rose. High correlation with changes in oncogene expression
