目的研究嗜酸乳杆菌DOMLa菌株是否对致泻性大肠埃希菌对上皮细胞粘附和侵袭能力具有抑制作用。方法使用了Hep-2细胞粘附实验和Hep-2细胞侵袭实验。结果发现嗜酸乳杆菌DOMLa菌株对Hep-2细胞粘附平均达222.48±30.7个细菌/细胞。thyA基因突变菌株DOMLaF84对Hep-2细胞的粘附为61.96±10.27个细菌/细胞。将嗜酸乳杆菌DOMLa菌株与肠产志贺氏样毒素且具有侵袭力的大肠埃希氏菌(ESIEC)F171菌株共同孵育后,可将后者的粘附和侵袭菌数分别由273.4±24.7个细菌/细胞和13.4±4.9个细菌/细胞,降低到65.3±13.1个细菌/细胞和6.82±1.6个细菌/细胞,差异有显著性(P<0.01)。将DOMLa菌株与肠侵袭性大肠埃希氏菌(EIEC)8401菌株共同孵育后,可将后者对Hep-2细胞的粘附和侵袭数分别由269.2±26.5个细菌/细胞和34.0±5.3个细菌/细胞降到70.7±14.0个细菌/细胞和6.82±2.9个细菌/细胞,差异有显著性(P<0.01)。结论嗜酸乳杆菌DOMLa菌株对ESIECF171菌株和EIEC8401菌株的Hep-2细胞的粘附和侵袭能力?
Objective To study whether Lactobacillus acidophilus DOMLa could inhibit the ability of diarrheal Escherichia coli to adhere to and invade epithelial cells. Methods Hep-2 cell adhesion assays and Hep-2 cell invasion assays were used. As a result, it was found that the Lactobacillus acidophilus DOMLa strain had an average of 222.48 ± 30.7 bacteria / cell on Hep-2 cell adhesion. The adhesion of thyA gene mutant strain DOMLaF84 to Hep-2 cells was 61.96 ± 10.27 bacteria / cell. After co-incubation of the L. acidophilus DOMLa strain with the enteric-producing shiga-like toxin-producing Escherichia coli (ESIEC) F171 strain, the number of adhesion and invasion of the latter strain increased from 273.4 ± 24.7 bacteria / cell and 13.4 ± 4.9 bacteria / cell, down to 65.3 ± 13.1 bacteria / cell and 6.82 ± 1.6 bacteria / cell, the difference was significant (P <0.01). Incubation of the DOMLa strain with the E. coli 8401 strain of enterotoxigenic Escherichia coli (EIEC) showed that the adhesion and invasion rates of the latter to Hep-2 cells were increased from 269.2 ± 26.5 bacteria / cell and 34.0 ± 5.3 bacteria / cell down to 70.7 ± 14.0 bacteria / cell and 6.82 ± 2.9 bacteria / cell, the difference was significant (P <0.01).
