目的研究DNCB诱导小鼠耳肿胀试验中小鼠耳肿胀程度与不同组织内IL-4和IFN-γ表达的相关性。方法以0.50%的DNCB溶液于小鼠背部诱导致敏3d,7d后测量小鼠双耳厚度并分组,以2.00%,1.00%,0.50%,0.25%,0.10%,0.05%和0七种浓度的DNCB溶液于各组小鼠左耳激发,右耳作为对照。每24h测量小鼠双耳厚度并于激发后72h处死小鼠,取血液、双侧耳组织、两侧耳部回流淋巴结,测定组织内IL-4和IFN-γ的表达。结果小鼠激发侧耳肿胀指数与DNCB激发浓度成线性正相关(P<0.01)。小鼠激发侧耳组织内IL-4高表达,并与激发后48h耳肿胀指数正相关,相关系数为0.78(P<0.01)。IFN-γ与激发后48h耳肿胀指数相关系数较低或不相关。结论小鼠耳肿胀程度与DNCB激发浓度、激发侧耳组织内IL-4存在相关性。
Objective To investigate the correlation between the degree of ear edema in mice induced by DNCB and the expression of IL-4 and IFN-γ in different tissues. Methods Sensitized mice were sensitized with DNCB solution of 0.50% for 3 days. After 7 days, the thickness of ears of mice was measured and divided into groups of 2.00%, 1.00%, 0.50%, 0.25%, 0.10%, 0.05% and 0 DNCB solution in each group mice left ear excitation, the right ear as a control. The thickness of the ears of mice was measured every 24h, and the mice were sacrificed at 72h after challenge. Blood, bilateral ear tissues and bilateral auricular reflux nodes were collected to determine the expression of IL-4 and IFN-γ in the tissues. Results The stimulated Pleurotus ostreatus index in mice was linearly correlated with the concentration of DNCB challenge (P <0.01). The expression of IL-4 in lateral ear tissues was significantly increased in mice, and positively correlated with ear swelling index at 48h after challenge. The correlation coefficient was 0.78 (P <0.01). The correlation coefficient between IFN-γ and ear swelling index at 48h after challenge was low or not correlated. Conclusion The degree of ear swelling in mice is correlated with the concentration of DNCB and the level of IL-4 in the stimulated lateral ear tissues.
