目的了解朗格汉斯细胞(LC)相关趋化因子基因在尖锐湿疣(CA)以及正常表皮中的表达。方法采用Affy-metrixHG-U133A2.0寡核苷酸芯片,对3例CA表皮和3例正常表皮LC相关趋化因子基因的表达进行了检测,并应用半定量RT-PCR验证其中部分基因的差异表达。结果基因芯片检测到CXCL2、CXCL8、CXCR4、CCL3/CCL3L1、CCL5、CCL20及CCL27共7个LC相关趋化因子基因在CA表皮中的表达普遍下调;半定量RT-PCR验证表明,CCL20和CXCR4基因在CA表皮中的表达显著下调。结论LC相关趋化因子基因在CA表皮中的表达普遍下调,这些基因表达的下调可能与CA表皮中LC数目减少和归巢障碍有关。
Objective To investigate the expression of Langerhans cell (LC) -related chemokines in condyloma acuminatum (CA) and normal epidermis. Methods Affy-metrixHG-U133A2.0 oligonucleotide microarray was used to detect the expression of LC-related chemokine genes in 3 cases of CA epidermis and 3 cases of normal epidermis. Semi-quantitative RT-PCR was used to verify the differences of some genes expression. Results The gene expression of CXCL2, CXCL8, CXCR4, CCL3 / CCL3L1, CCL5, CCL20 and CCL27 was downregulated in CA epidermis. The results of semi-quantitative RT-PCR showed that CCL20 and CXCR4 genes The expression in CA epidermis was significantly down-regulated. Conclusion The expression of LC-related chemokines is down-regulated in CA epidermis. The down-regulation of these genes may be related to the decrease of LC number in CA epidermis and homing disorders.
