目的比较缺氧对世居高原藏族人和移居高原汉族人脐静脉内皮细胞(UVECs)血管内皮生长因子(VEGF)、诱导型一氧化氮合酶(iNOS)和内皮型一氧化氮合酶(eNOS)mRNA表达的影响。方法分离脐静脉内皮细胞,体外原代培养,分为①世居高原藏族组和②移居高原汉族组两组,每组包括常氧对照和缺氧(0.5%O2)2h、4h、12h和24h时点。分离细胞总RNA,分别用RT-PCR检测VEGF、iNOS和eNOSmRNA表达。结果缺氧可以诱导世居高原藏族人UVECs表达iNOS和VEGFmRNA表达增加,同时抑制eNOSmRNA表达。移居高原汉族人UVECs表达上述基因mRNA的特点与世居高原藏族人相似。结论缺氧时世居高原藏族人UVECsVEGF、iNOS和eNOSmRNA表达特点与移居高原汉族人相似,提示上述基因表达的改变可能是缺氧反应机制的共同过程。
Objective To compare the effects of hypoxia on the expression of vascular endothelial growth factor (VEGF), inducible nitric oxide synthase (iNOS) and endothelial nitric oxide synthase (eNOS) in human umbilical vein endothelial cells (UVECs) ) mRNA expression. Methods Human umbilical vein endothelial cells were isolated and subcultured in vitro. The cells were divided into two groups: ① native Tibetan plateau group and ② migrant highland plateau group. Each group consisted of normoxia control and hypoxia (0.5% O2) for 2h, 4h, 12h and 24h Time. Total RNA was isolated and the expression of VEGF, iNOS and eNOS mRNA was detected by RT-PCR. Results Hypoxia could induce the expression of iNOS and VEGF mRNA in UVECs of Tibetan Plateau Tibet Autonomous Region and increase the expression of eNOS mRNA. The characteristics of mRNA expression of UVECs in Han immigrants in Plateau are similar to Tibetans living in Plateau. Conclusion The expressions of VEGF, iNOS and eNOSmRNA in the Tibetan Plateau in hypoxia are similar to those of Han Chinese who migrated to the plateau, indicating that the above gene expression may be the common process of hypoxia response.
