目的研究大肠癌细胞对化疗药物的敏感性与p53基因突变的相关性。方法用MTT比色法检测20位大肠癌患者的癌细胞对化疗药物的敏感性,并用PCR-SSCP技术分析其癌细胞的p53基因突变。结果大肠癌细胞对5-Fu[IR=(54.7±14.1)%]比对MMC[IR=(43.9±10.8)%]及ADM[IR=(43.9±9.4)%]更敏感(P<0.05),但对DDP有耐药性[IR=(25.9±12.8)%,P<0.01]。p53基因的突变率为45%(9/20)。癌细胞对MMC和ADM的敏感性与p53基因突变呈正相关(rmmc=0.589,radm=0.624,P<0.005)。结论化疗药物MMC和ADM对大肠癌的作用依赖于p53基因的状态,而大肠癌对5-Fu和DDP的敏感性却与p53基因突变无关。
Objective To study the correlation between chemosensitivity of colorectal cancer cells and p53 gene mutation. Methods MTT assay was used to detect the sensitivity of cancer cells to chemotherapeutics in 20 patients with colorectal cancer. The mutation of p53 gene in cancer cells was analyzed by PCR-SSCP. Results Colorectal cancer cells were more sensitive to 5-Fu (54.7 ± 14.1%) than MMC (43.9 ± 10.8%) and ADM (43.9 ± 9.4% , But resistant to DDP [IR = (25.9 ± 12.8)%, P <0.01]. The mutation rate of p53 gene was 45% (9/20). The sensitivity of cancer cells to MMC and ADM was positively correlated with p53 gene mutation (rmmc = 0.589, radm = 0.624, P <0.005). Conclusion The effect of chemotherapy drugs MMC and ADM on colorectal cancer depends on the status of p53 gene. However, the sensitivity of colorectal carcinoma to 5-Fu and DDP is not related to the mutation of p53 gene.
