目的:探讨克服肿瘤细胞多药耐药(MDR)的方法,提高化疗效果。方法:采用多药耐药反义基因(mdr-1-ASPS-ODN)逆转K562/ADM肿瘤细胞的MDR,而诱导肿瘤细胞凋亡。结果:mdr-1-ASPS-ODN诱导K562/ADM细胞产生大量DNA断片,流式细胞仪检测发现几乎全部mdr-1+K562/ADM细胞发生凋亡。结论:mdr-1-ASPS-ODN能有效、特异地抑制mdr-1基因表达,逆转肿瘤细胞的MDR,促进阿霉素诱导K562/ADM细胞凋亡,为其临床应用提供理论依据。
Objective: To explore ways to overcome the multidrug resistance (MDR) of tumor cells and improve the efficacy of chemotherapy. METHODS: Multidrug resistance antisense gene (mdr-1-ASPS-ODN) was used to reverse the MDR of K562/ADM tumor cells and induce tumor cell apoptosis. RESULTS: mdr-1-ASPS-ODN induced large numbers of DNA fragments in K562/ADM cells. Flow cytometry revealed that almost all mdr-1+K562/ADM cells developed apoptosis. Conclusion: mdr-1-ASPS-ODN can effectively and specifically inhibit the expression of mdr-1 gene, reverse the MDR of tumor cells, and promote apoptosis of K562/ADM cells induced by doxorubicin, which provides a theoretical basis for its clinical application.
