目的探讨邻苯二甲酸二丁酯(DBP)对青春期雌性大鼠卵巢PPARγ mRNA表达的影响。方法将32只健康21日龄SPF级SD雌性大鼠按体重随机分为对照(玉米油)组和低(250 mg/kg)、中(500 mg/kg)、高剂量(1 000 mg/kg)DBP染毒组,每组8只。采用灌胃方式进行染毒,染毒容积为5 ml/kg,连续染毒8周。在动情前期处死大鼠,采用RT-PCR法检测大鼠卵巢组织过氧化物酶体增生物激活受体-γ(peroxisome proliferator-activated receptor-γ,PPARγ)mRNA的表达水平。结果与对照组比较,500及1 000 mg/kg DBP染毒组大鼠卵巢PPARγmRNA表达水平升高,差异有统计学意义(P<0.05)。且随着DBP染毒剂量的升高,大鼠卵巢PPARγmRNA的表达水平呈上升趋势。结论 DBP有可能通过激活卵巢PPARγmRNA表达而对雌性大鼠产生生殖毒性。
Objective To investigate the effect of dibutyl phthalate (DBP) on ovarian PPARγ mRNA expression in adolescent female rats. Methods 32 healthy SDF female SD rats aged 21 days were randomly divided into control (corn oil) group and low (250 mg / kg), medium (500 mg / kg) and high dose ) DBP exposure group, 8 in each group. Gavage by way of exposure, the volume of 5 ml / kg, continuous exposure for 8 weeks. The rats were sacrificed at the early estrus stage, and the mRNA expression of peroxisome proliferator-activated receptor-γ (PPARγ) in ovary tissue of rats was detected by RT-PCR. Results Compared with the control group, the levels of PPARγmRNA in the ovaries of 500 and 1 000 mg / kg DBP groups were significantly increased (P <0.05). And with the increase of DBP dose, the expression level of PPARγmRNA in rat ovary increased. Conclusion DBP may cause reproductive toxicity in female rats by activating ovarian PPARγ mRNA expression.
