目的探讨血管紧张素Ⅱ(angiotensin Ⅱ,AngⅡ)对前脂肪细胞分化过程中转录因子PPARγ、C/EBPα、SREBP1-c/ADD-1表达的调节从而研究AngⅡ调节前脂肪细胞分化可能的分子机制。方法 3T3-L1前脂肪细胞体外传代培养及诱导分化,用RT-PCR技术检测诱导分化过程中不同浓度AngⅡ处理6d对前脂肪细胞分化相关转录因子PPARγ、C/EBPα、SREBP1-c/ADD-1 mRNA表达水平的影响。结果 RT-PCR结果显示,100 nmol/L的AngⅡ处理细胞6 d后,PPARγ、C/EBPα、SREBP1-c/ADD-1 mRNA的相对表达量与对照组(AngⅡ添加浓度为0 nmol/L)有显著性差异(P<0.05),比对照组相应的转录因子分别增加了48%、50%、43%。结论前脂肪细胞分化过程中AngII能够上调PPARγ、C/EBPα、SREBP1-c/ADD-1 mRNA的表达量,AngⅡ可能通过影响PPARγ、C/EBPα和SREBP1-c/ADD-1的表达来调节前脂肪细胞的分化过程。
Objective To investigate the regulation of angiotensin Ⅱ (AngⅡ) on the expression of transcription factors PPARγ, C / EBPα and SREBP1-c / ADD-1 during the differentiation of preadipocytes in order to study the possible molecular mechanism of angiotensin Ⅱ (AngⅡ) regulating preadipocyte differentiation. Methods 3T3-L1 preadipocytes were subcultured and differentiated in vitro. The expression of PPARγ, C / EBPα, SREBP1-c / ADD-1 in preadipocytes were detected by RT-PCR at different concentrations of AngⅡ for 6 days mRNA expression levels. Results The RT-PCR results showed that the relative expression of PPARγ, C / EBPα and SREBP1-c / ADD-1 mRNA in AngⅡ-treated cells at 100 nmol / L for 6 d was significantly higher than that in the control group (P <0.05). Compared with the control group, the corresponding transcription factors increased by 48%, 50% and 43% respectively. Conclusions AngII can up-regulate the mRNA expression of PPARγ, C / EBPα and SREBP1-c / ADD-1 in the process of preadipocyte differentiation. AngⅡ may regulate the expression of PPARγ, C / EBPα and SREBP1-c / Adipocyte differentiation process.
