目的探讨IL-10、TNF-α对孕鼠胎盘生乳素(rPL)的影响。方法将孕鼠随机分为6组。第1组为生理盐水组,第2组为20ng TNF-α组,第3组为50ng IL-10组,第4组为20ng TNF-α+50ng IL-10组,第5组为20ng TNF-α+100ng IL-10组,第6组为20ng TNF-α+200ng IL-10组。分别于妊娠第7、8、9、10、11d给第1组腹腔注射生理盐水,其余5组腹腔注射不同量的IL-10、TNF-α。于妊娠第12d用ELISA法测定6组小鼠血清中rPL的水平。用方差分析及秩和检验比较6组小鼠血清rPL的含量。结果 1.第2组的血清rPL水平比第1组及第3组明显升高,差异有显著性(P<0.01);第3组血清rPL水平虽比第1组升高,但两组数据无显著性差异(P>0.05);2.第4组、第5组和第6组血清rPL水平均低于第2组,4组数据差异无统计学意义(P>0.05)。结论大量的TNF-α可显著升高孕鼠血清中胎盘生乳素的水平。
Objective To investigate the effect of IL-10 and TNF-α on placental lactogen (rPL) in pregnant rats. Methods Pregnant mice were randomly divided into 6 groups. The first group was saline group, the second group was 20ng TNF-α group, the third group was 50ng IL-10 group, the fourth group was 20ng TNF-α + 50ng IL-10 group, the fifth group was 20ng TNF- α + 100ng IL-10 group, group 6 was 20ng TNF-α + 200ng IL-10 group. The first group was given intraperitoneal injection of saline on the 7th, 8th, 9th, 10th, and 11th day respectively. The other 5 groups were injected intraperitoneally with different doses of IL-10 and TNF-α. The serum levels of rPL in 6 groups of mice were measured by ELISA method on the 12th day of gestation. The levels of serum rPL in six groups of mice were compared by ANOVA and rank sum test. The level of serum rPL in group 2 was significantly higher than that in group 1 and group 3 (P <0.01), while the level of serum rPL in group 3 was higher than that in group 1 There was no significant difference (P> 0.05) .2. The serum rPL levels in group 4, group 5 and group 6 were lower than those in group 2, and there was no significant difference in group 4 (P> 0.05). Conclusions A large number of TNF-α can significantly increase the level of placental lactogen in pregnant rats.
