目的:观察百草枯对PC12细胞的活化素不同亚基和受体表达的影响。方法:将百草枯加入体外培养的PC12细胞中,用RTPCR法检测细胞的活化素βAmRNA、βBmRNA、抑制素αmRNA和受体ⅡAmRNA表达水平的变化,并以锥虫蓝排斥法检测细胞活力的变化。结果:百草枯损害后PC12细胞的活化素βAmRNA、βBmRNA和受体ⅡAmRNA表达水平明显下调,抑制素αmRNA的表达水平无明显变化。细胞活力在百草枯损害后12和24h下降。结论:百草枯可能通过引起PC12细胞的活化素和受体表达水平的下调,而导致细胞的损害。
Objective: To observe the effect of paraquat on the expression of different subunits and receptors of activin in PC12 cells. Methods: Paraquat was added into PC12 cells cultured in vitro. The expression of activin βA mRNA, βB mRNA, inhibin α mRNA and receptor ⅡA mRNA were detected by RTPCR. The changes of cell viability were detected by trypan blue exclusion assay. Results: The expressions of activin βA mRNA, βB mRNA and receptor ⅡA mRNA in PC12 cells were significantly down-regulated after paraquat-induced injury, while the expression of inhibin α mRNA did not change significantly. Cell viability decreased at 12 and 24h after paraquat damage. Conclusion: Paraquat may cause cell damage by down-regulation of activin and receptor expression in PC12 cells.
