目的:观察桃红四物汤对大鼠创伤性股骨头缺血性坏死进程中GPR48表达的影响,探讨桃红四物汤改善坏死股骨头血运及坏死骨新生的作用效果。方法:大鼠造模成功后随机分为桃红四物汤低、中、高剂量组、模型组。桃红四物汤组采用低、中、高剂量灌胃,正常对照组、模型组采用等量的生理盐水灌胃,灌胃8周后收集样本,观察样本大体外观、组织形态学、空骨陷窝率;RT-PCR检测大鼠股骨头坏死过程中GPR48的表达情况。结果:(1)大体样本观察:模型组大鼠股骨头呈暗红色,粗糙,有不同程度的关节软骨面塌陷。桃红四物汤组与模型组基本一致,但塌陷程度较轻。(2)X线结果:模型组、桃红四物汤组出现密度减低区,骨小梁排列紊乱,有不同程度的关节软骨面塌陷。(3)HE染色:模型组、桃红四物汤组股骨头髓腔空虚,造血组织、骨小梁骨细胞大量减少,空骨陷窝大量形成。(4)RT-PCR分析结果:正常对照组、桃红四物汤组、模型组的GPR48表达量逐渐减少,但桃红四物汤组不同剂量之间,随着剂量增高GPR48表达量逐渐升高,差异均有统计学意义(P<0.01)。结论:桃红四物汤可促进大鼠创伤性股骨头缺血性坏死进程中GPR48的表达。
Objective: To observe the effect of Taohongsiwutang on the expression of GPR48 during the process of traumatic avascular necrosis of the femoral head in rats, and to explore the effect of Taohongsiwutang on improving the necrotic femoral head blood circulation and the necrotic bone newborn. Methods: After successful modeling, rats were randomly divided into low, medium and high dose Taohongsiwutang, model group. The rats in Taohong Siwu Decoction group were given low, medium and high doses of gavage. The normal control group and model group were given gavage with normal saline. After 8 weeks of gavage, the samples were collected to observe the general appearance, histomorphology, Fossa rate; RT-PCR detection of rat head necrosis of the expression of GPR48. Results: (1) The general sample observation: the model group rats femoral head was dark red, rough, with varying degrees of articular cartilage collapse. Taohong Siwu Decoction group and the model group basically the same, but the collapse of the lighter. (2) X-ray results: In the model group and Taohong Siwu Decoction group, the densities were reduced and the trabeculae were disordered, and the cartilage surface collapsed to varying degrees. (3) HE staining: In the model group and the Taohong Siwu Decoction group, the femoral head cavity was empty, the hematopoietic tissue and trabecular bone cells were greatly reduced, and the empty bone lacuna was formed in a large amount. (4) The result of RT-PCR showed that the expression of GPR48 in normal control group, Taohongsiwutang group and model group decreased gradually, but the expression of GPR48 in Taohong Siwu Decoction group increased gradually with the increase of dose, The differences were statistically significant (P <0.01). Conclusion: Taohong Siwu Decoction can promote the expression of GPR48 during traumatic avascular necrosis of the femoral head in rats.
