目的通过导向分离法发现姜黄Curcuma longa根茎中1个新化合物,并进行分离鉴定及细胞毒活性研究。方法运用LC-MS分析技术,结合MCI-Gel小孔树脂、ODS-C18反相柱及RP-HPLC等柱色谱手段分离纯化,根据理化性质和波谱数据鉴定化合物的结构。结果从姜黄根茎甲醇提取物的低极性流分中分离得到1个姜黄素类新化合物,鉴定为姜黄素-没药烷型倍半萜二聚体化合物3,4′′′-epoxy-5′′′-C-(1α,2β,3β-bisabola-4,10-diene-9-one)-(2→5′′′)-curcumin(1),该化合物对Hep G2、A549和CT-26肿瘤细胞IC50值分别为4.82、8.81和5.40μmol/L。结论化合物1为1个姜黄素类新化合物,命名为姜萜素G;其对肿瘤细胞具有显著的细胞毒活性。
OBJECTIVE: To find out a new compound in Curcuma longa rhizome by directed separation method, and to isolate and identify and study its cytotoxicity. Methods The compounds were separated and purified by LC-MS, MCI-Gel resin, ODS-C18 and RP-HPLC. The structures of compounds were identified by their physicochemical properties and spectral data. Results A new curcuminoid compound was isolated from the low polarity fractions of methanol extract of turmeric rhizome and was identified as curcumin - myrothecyl type sesquiterpene dimer compound 3,4 ’"- epoxy - 5 (2 → 5 ’’ ’) - curcumin (1), which is effective against Hep G2, A549 and CT- 26 tumor cells with IC50 values of 4.82, 8.81 and 5.40 μmol / L, respectively. Conclusion Compound 1 is a new curcuminoid compound named as zingiberin G; it has significant cytotoxic activity on tumor cells.
