目的探讨人NHE1反义基因转染对SGC-7901胃癌细胞株增殖和凋亡的影响,探索胃癌治疗的新方法。方法采用脂质体法将构建好的人NHE1基因反义真核表达载体转染至SGC-7901胃癌细胞中,比较观察细胞转染前后生长曲线、双层软琼脂克隆形成能力、裸鼠成瘤能力以及细胞周期和细胞凋亡率的变化。结果NHE1反义基因能使SGC-7901胃癌细胞生长速度减慢,双层软琼脂集落形成能力、体外生长增殖能力降低,细胞凋亡率增加,裸鼠体内成瘤能力显著降低。结论反义NHE1基因能抑制SGC-7901胃癌细胞增殖,诱导细胞凋亡,具有良好的胃癌治疗效果,提示NHE1基因可成为胃癌治疗的新靶点,具有一定的临床应用前景。
Objective To investigate the effect of human NHE1 antisense gene transfection on the proliferation and apoptosis of gastric cancer cell line SGC-7901 and to explore a new method of gastric cancer treatment. Methods The antisense eukaryotic expression vector of human NHE1 gene was constructed and transfected into SGC-7901 gastric cancer cells by lipofectamine. The growth curve, double soft agar colony forming ability and nude mice tumorigenesis Ability and change of cell cycle and apoptosis rate. Results The antisense gene of NHE1 could slow down the growth of SGC-7901 gastric cancer cells, reduce the growth of double soft agar, reduce the growth and proliferation ability, increase the apoptosis rate of SGC-7901 gastric cancer cells. Conclusion The antisense NHE1 gene can inhibit the proliferation and induce apoptosis of gastric cancer cell line SGC-7901 and has a good therapeutic effect on gastric cancer. It suggests that NHE1 gene may be a new therapeutic target for gastric cancer and has certain clinical application prospects.
