目的:检测转化生长因子βⅠ(TGFβⅠ)、转化生长因子βⅡ受体(TGFβRⅡ)和细胞周期素E(cyclin E)蛋白在尖锐湿疣(CA)组织中的表达。方法:采用免疫组化SP法和图像分析系统检测30例CA患者皮损,15名正常人皮肤(包皮)组织中TGFβⅠ、TGFβRⅡ及cyclin E的表达。结果:TGFβI和cyclin E在CA组织中的表达均显著高于正常皮肤组织(P<0.05);TGFβRⅡ在CA组织中的表达显著低于正常皮肤组织(P<0.05);TGFβⅠ与cyclin E在CA组织中的表达呈正相关(P<0.05);TGFβRⅡ与cyclin E在CA组织中的表达呈负相关(P<0.05)。结论:TGFβRⅡ表达下调,使TGFβ信号传导通路受阻,降低了TGFβⅠ对cyclin E表达的抑制作用,及其对细胞增殖的负性调节作用,使CA组织过度增殖,可能是CA发病机制之一。
Objective: To detect the expression of TGFβⅠ, TGFβRⅡ and cyclin E in condyloma acuminatum (CA). Methods: The expressions of TGFβⅠ, TGFβRⅡ and cyclin E in the skin of 30 cases of CA and the normal skin (foreskin) were detected by immunohistochemical SP method and image analysis system. Results: The expressions of TGFβI and cyclin E in CA tissues were significantly higher than those in normal skin tissues (P <0.05). The expression of TGFβRII in CA tissues was significantly lower than that in normal skin tissues (P <0.05) There was a positive correlation between the expression of TGFβRⅡ and cyclin E in CA tissue (P <0.05). CONCLUSION: The down-regulation of TGFβRⅡ inhibits the TGFβ signaling pathway, reduces the inhibitory effect of TGFβⅠ on the expression of cyclin E, and negatively regulates the proliferation of the cells. The over-proliferation of CA tissue may be one of the pathogenesis of CA.
