应用cDNA阵列技术寻找遗传性癫差异表达基因 ,为从基因水平探讨癫的发病机理打下基础。采用AtlasTM RatcDNAExpressionArray建立遗传性癫易感性P77PMC大鼠和正常Wistar大鼠大脑皮质基因表达谱 ,用图像分析仪分析两者基因表达谱差异。结果发现有 15个差异表达基因 ,其中 13个基因在P77PMC大鼠大脑皮质中高表达而在正常Wistar大鼠大脑皮质中低表达 ,2个基因在正常Wistar大鼠大脑皮质中高表达而在P77PMC大鼠大脑皮质中低表达。结果提示 ,P77PMC大鼠与正常Wistar大鼠大脑皮质存在多个差异表达基因 ,这些差异表达的基因可能在癫的发生、发展中扮演了重要角色。
The use of cDNA array technology to find the genetically-modified epilepsy differentially expressed genes, to lay the foundation for exploring the pathogenesis of epilepsy at the gene level. AtlasTM RatcDNAExpressionArray was used to establish the gene expression profiles of cerebral cortex in hereditary epilepsy susceptible P77PMC rats and normal Wistar rats. The difference of gene expression profiles between the two groups was analyzed by image analyzer. Fifteen differentially expressed genes were found. Thirteen of them were highly expressed in cerebral cortex of P77PMC rats, but low in normal cerebral cortex of Wistar rats. Two of them were highly expressed in the cerebral cortex of normal Wistar rats. However, in P77PMC rats Low expression in the cerebral cortex. The results suggest that there are many differentially expressed genes in the cerebral cortex of P77PMC and normal Wistar rats. These differentially expressed genes may play an important role in the occurrence and development of epilepsy.
