液体深层培养猪苓多糖脱色技术的研究

目的研究液体深层培养猪苓菌丝体水提部位粗多糖脱色的最佳工艺条件。方法采用单因素试验和正交试验分别考察双氧水、活性炭、DEAE纤维素等3种脱色剂对菌丝体粗多糖的脱色效果,用光谱扫描法检测光吸收强度的变化。结果活性炭作为脱色剂,效果优于其他2种,脱色最佳工艺条件为:温度60℃,时间40min,活性炭添加量3g·100mL-1。结论实验中优化后的脱色方法简便、易行,也为猪苓菌丝体多糖的工业化提取中脱色工序的操作提供理论依据。 Objective To study the optimum conditions for the crude polysaccharide decolorization of the mycelium of Polyporus umbellatus cultured in liquid. Methods The single factor experiment and orthogonal experiment were used to study the decolorization effect of three kinds of decoloring agents such as hydrogen peroxide, activated carbon and DEAE cellulose on the mycelium crude polysaccharide. The changes of light absorption intensity were detected by spectral scanning. Results As the decoloring agent, activated carbon was better than the other two kinds of decoloring agents. The optimal decoloration conditions were as follows: temperature 60 ℃, time 40min, amount of activated carbon 3g · 100mL-1. Conclusion The optimized decolorization method in the experiment is simple and easy to operate, and provides a theoretical basis for the operation of decolorization in the industrial extraction of Polyporus umbellatus polysaccharide.
