测定多聚核糖体含量及多聚程度的分布在研究生物组织的生理生化现象和代谢过程中有重要意义。其定量分离的报道很多。本文利用新型离心转头、通过变更密度梯度配比,离心时间和离心速度3个参量,经过多次实验,获得一个新的制备方法,兹将结果介绍如下。先采用Dupont/Soruall TH641水平转头,离心管加入40%~10%线性蔗糖梯度,再加入粗制鼠肝多聚核糖体,温度10℃,转速3500r/min,离90min。再采用日立scp-85H超速离心机的RPV-50T垂直转头,40PA离心管,管底加20mol/L ST KM,其上是55%~15%
