β-catenin是Wnt信号传导通路和上皮钙黏连素-连环素复合体(E-cadherin/catenin)的关键分子。目前,在许多肿瘤研究中已发现β-catenin的异常蓄积,而且,其异常蓄积的程度与肿瘤发生、发展、浸润、转移相关。在正常上皮细胞中,β-catenin与E-cadherin相连,介导细胞间的黏附,而少量游离的β-catenin与大肠腺瘤息肉样蛋白(adenomatouspolyposis coli,APC)复合体结合,经泛素-蛋白酶小体系统降解。当Wnt信号激活时,细胞核内蓄积的β-catenin与TCF/LEF(Tcell factor/lymp hoid enhancer facor)结合,激活c-myc和cyclin D1等基因,调节细胞增殖。笔者就其与鳞癌发生、发展、浸润、转移至肩的关系方面的研究做一综述。
β-catenin is a key molecule of Wnt signaling pathway and E-cadherin / catenin. At present, abnormal accumulation of β-catenin has been found in many tumor studies. Moreover, the extent of abnormal accumulation is related to tumorigenesis, development, invasion and metastasis. In normal epithelial cells, β-catenin is connected with E-cadherin, which mediates cell adhesion. A small amount of free β-catenin binds with the complex of adenomatous polyposis coli (APC) Proteasome system is degraded. When Wnt signaling is activated, β-catenin accumulated in the nucleus binds to TCF / LEF (Tcell factor / lymphoid enhancer facor) and activates genes such as c-myc and cyclin D1 to regulate cell proliferation.
