初步探讨了栎叶枇杷胚培养获得愈伤组织,从胚愈伤组织分离原生质体以及原生质体的早期培养方法,着重研究了酶混合液对栎叶枇杷原生质体分离和培养的效应.结果表明:以1%纤维素酶和2%果胶酶配合使用效果最好,得率和成活率分别为(9.8±0.03)×10 ̄6和96.0%.作为酶混合液中渗透压稳定剂的甘露醇浓度则以11%-12%为好.从合适的酶混合液中得到的原生质体培养已获得愈伤组织.
The initial culture of callus from embryo culture of Quercus variabilis, culture of callus from embryo callus, and protoplast culture were studied. The effects of enzyme mixture on protoplast isolation and culture of Quercus lugens were also studied. The results showed that the best combination of 1% cellulase and 2% pectinase was obtained. The yield and survival rate were (9.8 ± 0.03) × 10 ~ 6 and 96.0%, respectively. The mannitol concentration as the osmotic pressure stabilizer in the enzyme mixture is preferably 11% to 12%. Callus has been obtained from protoplast culture obtained from a suitable enzyme mix.
