目的:研究miRNA-126基因敲减小鼠肠系膜淋巴结T及B淋巴细胞数量的变化。方法:检测miRNA-126基因敲减(KD)小鼠肠系膜淋巴结的体积、重量和淋巴结细胞总数;HE染色检测小鼠肠系膜淋巴结的病理形态学变化;流式细胞术(FACS)检测淋巴结中T及B淋巴细胞数量的变化。结果:与野生型(WT)小鼠相比,miR-126KD小鼠肠系膜淋巴结体积、重量和淋巴结细胞总数均显著增加(P<0.05),且病理形态学发生异常;肠系膜淋巴结中B淋巴细胞比例和数量无变化(P>0.05),CD4+T细胞数量无变化,而CD4+T细胞百分数明显减少(P<0.05),CD8+T细胞百分数和绝对数量均显著增加(P<0.05)。结论:miRNA-126敲减后可显著影响肠系膜淋巴结中T淋巴细胞的数量,提示miRNA-126可能影响肠系膜淋巴结的免疫功能。
Objective: To study the changes of T and B lymphocytes in mesenteric lymph node of miRNA-126 knockout mice. Methods: The mesenteric lymph node volume, weight and total number of lymph node in miRNA-126 knockout (KD) mice were detected. The pathological changes of mesenteric lymph nodes in mice were detected by HE staining. T lymphocyte subsets and lymph node metastasis were detected by flow cytometry (FACS) B lymphocyte number changes. Results: Compared with WT mice, miR-126KD mice had significantly increased mesenteric lymph node volume, weight and total number of lymph node cells (P <0.05), as well as abnormal pathomorphology. The proportion of B lymphocytes in mesenteric lymph nodes (P <0.05). There was no change in the number of CD4 + T cells, but the percentage of CD4 + T cells was significantly decreased (P <0.05), and the percentage and absolute number of CD8 + T cells were significantly increased (P <0.05). Conclusion: The knockdown of miRNA-126 can significantly affect the number of T lymphocytes in mesenteric lymph nodes, suggesting that miRNA-126 may affect the immune function of mesenteric lymph nodes.
