目的观察体外培养的HT-29细胞在丁酸钠作用后形态学及生长曲线的变化。初步探讨丁酸钠对结肠癌细胞的凋亡诱导作用。方法利用相差显微镜、可变投射显微镜、电镜观察细胞形态学变化,MTT法检测HT-29细胞生长情况。结果丁酸钠作用后的HT-29细胞形态发生改变,表现为细胞体积变小、形状变圆,细胞核固缩,脱壁细胞逐渐增多,细胞膜突起减少,核内染色质呈团块状积聚于核膜下,并可见凋亡小体。丁酸钠2.5、5和10mM对HT-29细胞生长均有抑制作用,并呈浓度、时间依赖性。结论细胞形态学的变化初步显示了丁酸钠对结肠癌细胞具有凋亡诱导作用。
Objective To observe the morphological changes and growth curves of HT-29 cells incubated with sodium butyrate in vitro. Preliminary study of sodium butyrate on colon cancer cell apoptosis induced. Methods The morphological changes of cells were observed by phase-contrast microscope, variable projection microscope and electron microscope. The growth of HT-29 cells was detected by MTT assay. Results The morphological changes of HT-29 cells induced by sodium butyrate showed that the cell volume became smaller, the shape of the cells became round, the nuclei were pyknotic, the number of detached cells increased gradually, the number of cell membrane protuberances decreased, Nuclear membrane, and apoptotic bodies can be seen. Sodium butyrate 2.5, 5 and 10 mM inhibited the growth of HT-29 cells in a concentration-and time-dependent manner. Conclusion The morphological changes of the cells initially showed that sodium butyrate has an apoptosis-inducing effect on colon cancer cells.
