目的:利用响应曲面分析法优化毛果鱼藤总生物碱的提取工艺,并检测其抑菌活性。方法:以毛果鱼藤为原料,采用微波提取法,设计单因素实验,分别考察了料液比、乙醇体积分数、提取时间、微波提取功率、提取次数5个单因素对毛果鱼藤总生物碱提取效果的影响;采用响应曲面法优化毛果鱼藤总生物碱的提取工艺,提高总生物碱的提取效率,并考察总生物碱的体外抑菌作用。结果:毛果鱼藤总生物碱的最佳提取工艺条件为微波功率400 W,提取温度为70℃,料液比为1∶23,乙醇体积分数为70%,提取时间为23 min,在此条件下,以盐酸小檗碱计,总生物碱提取率为0.107 7 mg·g-1;抑菌实验表明,毛果鱼藤总生物碱对金黄色葡萄球菌有一定的抑菌作用。结论:微波法提取生物碱工艺方法科学、稳定,且生物碱具有良好的抑菌作用。
OBJECTIVE: To optimize the extraction process of total alkaloids from Rhinophangium villosum by response surface methodology and to test its antibacterial activity. Methods: Single-factor experiment was carried out by using microwave-assisted extraction and microwave extraction method. The effects of solid-liquid ratio, ethanol volume fraction, extraction time, microwave extraction power and extraction times were investigated. Alkaloids extraction. The response surface methodology was used to optimize the extraction of total alkaloids and increase the total alkaloid extraction efficiency. The antibacterial activity of total alkaloids in vitro was also investigated. Results: The optimal extraction conditions of total alkaloids of P. sinica were microwave power of 400 W, extraction temperature of 70 ℃, solid-liquid ratio of 1:23, ethanol concentration of 70% and extraction time of 23 min, Under the conditions of berberine hydrochloride, the total alkaloid extraction rate of 0.107 7 mg · g-1; antibacterial experiments showed that the fruit Virgin saponin alkaloids Staphylococcus aureus have some antibacterial activity. Conclusion: Microwave extraction of alkaloids process method is scientific and stable, and alkaloids have good antibacterial activity.
