[目的]分析裸鼠皮下人鼻咽癌移植瘤模型体内血清中的蛋白指纹图谱,并与正常对照裸鼠比较,筛选差异表达蛋白,为鼻烟癌的早期诊断提供特有的血清蛋白标志物。[方法]采用表面增强激光解吸电离飞行时间质谱(SELDI-TOF-MS)蛋白芯片技术检测12只正常裸鼠和45只裸鼠皮下人鼻咽癌移植瘤模型体内血清中的蛋白指纹图谱,用Ciphergen ProteinChip 3.0软件进行数据的校正与分析,采用Ciphergen Biomaker Wizard 3.1版软件分析筛选对照与试验组间的差异表达蛋白。[结果]对照组与试验组间检出17个差异表达的蛋白质(P﹤0.0001),其质荷比(M/Z)分别为13447、14586、8948、8017、5553、6742、14743、11586、15675、9178、6423、14940、15492、11528、8615、6738、8102。分子量主要集中在20000D以内,大多数标志蛋白在试验组中表达明显增高,少数为低表达。在裸鼠皮下人鼻咽癌移植瘤模型体内随时间的推移而出现一些规律性表达的蛋白质,质荷比(M/Z)为6738、8017、8102、8615的4个蛋白质在接种癌细胞后表达逐渐增强,质荷比(M/Z)为14740、15484的两个蛋白质表达逐渐减弱。[结论]在正常裸鼠和裸鼠皮下人鼻咽癌移植瘤模型之间发现具有一定价值的差异表达蛋白,对鼻咽癌早期诊断的微量蛋白标志物的研究具有极其重要的意义。
[Objective] The purpose of this study was to analyze the protein fingerprints of in vivo serum of human nasopharyngeal carcinoma xenografts in nude mice and to screen differentially expressed proteins compared with normal control nude mice to provide specific serum protein markers for nasopharyngeal carcinoma. [Method] The protein fingerprint of sera from 12 normal and 45 nude mice bearing human nasopharyngeal carcinoma xenografts was detected by SELDI-TOF-MS. Ciphergen ProteinChip 3.0 software for data calibration and analysis, using Ciphergen Biomaker Wizard version 3.1 software analysis of screening control and experimental groups of differentially expressed proteins. [Results] A total of 17 differentially expressed proteins (P <0.0001) were detected between the control group and the experimental group, and their mass-to-charge ratios (M / Z) were 13447, 14586, 8948, 8017, 5553,6742, 14743, 11586, 15675,9178,6423,14940,15492,11528,8615,6738,8102. The molecular weight mainly concentrated in the 20000D or less, most of the marker protein was significantly increased in the experimental group, a small number of low expression. In the subcutaneous human nasopharyngeal carcinoma xenograft model in nude mice with the passage of time there are some regular expression of the protein, the mass-to-charge ratio (M / Z) of 6738,8017,8102,8615 four proteins after inoculation of cancer cells The expression gradually increased. The expression of two proteins with mass / charge ratio (M / Z) of 14740 and 15484 gradually decreased. [Conclusion] It is of great importance to study the differential diagnosis of nasopharyngeal carcinoma nasopharyngeal carcinoma xenografts in nude mice and normal nude mice.
