导痰汤对颈动脉硬化家兔内皮细胞黏附分子-1及p21表达的影响

目的:研究导痰汤是否能通过对内皮细胞黏附分子-1(intercellular adhesion molecular-1,ICAM-1)和p21通路的调节而干预颈动脉粥样硬化。方法:将25只雄性新西兰白兔随机分为正常对照组、模型组、导痰汤治疗组(分为高、中、低剂量组)。对照组给予基础饲料喂饲,模型组和导痰汤治疗组予以高脂饲料喂饲,并在喂饲1周后实施颈动脉内膜空气干燥术,术后导痰汤治疗组分别按照10,8,6 g.kg-1,ig每日给药1次。4周后处死家兔,取每只家兔右侧颈动脉远端1 cm,固定后行HE染色,观察病理改变。应用免疫组化SABC法检测ICAM-1和p21表达情况。结果:模型组血管内皮细胞、平滑肌细胞的胞浆中有ICAM-1(0.200±0.035)和p21(0.297±0.097)强阳性表达,显著高于对照组ICAM-1(0.037±0.005),p21(0.078±0.016),(P<0.01)。导痰汤低、中、高剂量治疗组细胞内ICAM-1(0.166±0.043,0.104±0.011,0.098±0.010)和p21(中剂量:0.173±0.021,高剂量:0.128±0.026)阳性表达显著低于模型组(P<0.05),且随剂量的增加,导痰汤对ICAM-1和p21的抑制逐渐增强;而p21与ICAM-1水平呈显著正相关(r=0.772,P<0.01)。结论:导痰汤可以有效抑制颈动脉硬化家兔血管内皮细胞中ICAM-1和p21的表达。 Objective: To investigate whether Dantan Soup can interfere with carotid atherosclerosis by regulating the expression of intercellular adhesion molecule-1 (ICAM-1) and p21. Methods: Twenty-five male New Zealand white rabbits were randomly divided into normal control group, model group and Dantan decoction group (divided into high, medium and low dose groups). The control group was fed with basal diet, the model group and the YinTanTang treatment group were fed with high-fat diet, and carotid intima-air-drying was performed after 1 week of feeding. After the operation, 8,6 g.kg-1, ig administered once daily. After 4 weeks, the rabbits were sacrificed and the distal carotid artery of each rabbit was placed 1 cm distal to each rabbit. HE staining was performed to observe the pathological changes. The immunohistochemical SABC method was used to detect the expression of ICAM-1 and p21. Results: The positive expression of ICAM-1 (0.200 ± 0.035) and p21 (0.297 ± 0.097) in the cytoplasm of vascular endothelial cells and smooth muscle cells of the model group was significantly higher than that of ICAM-1 (0.037 ± 0.005), p21 0.078 ± 0.016), (P <0.01). The expression of ICAM-1 (0.166 ± 0.043,0.104 ± 0.011,0.098 ± 0.010) and p21 (medium dose: 0.173 ± 0.021, high dose: 0.128 ± 0.026) in the low, medium and high dose treatment groups of Guntan decoction group were significantly lower (P <0.05). With the increase of dose, YinTan Decoction could inhibit ICAM-1 and p21 gradually, while p21 and ICAM-1 had a significant positive correlation (r = 0.772, P <0.01). Conclusion: Dantan Decoction can effectively inhibit the expression of ICAM-1 and p21 in rabbit vascular endothelial cells of carotid artery atherosclerosis.
