目的进一步评价丹参酮Ⅱ_A纳米结构脂质载体(TⅡ_A-NLC)的性质,并考察其对HaCaT细胞增殖的影响。方法采用纳米粒度仪及HPLC法对TⅡ_A-NLC的粒径和光稳定性进行考察,透析袋法测定TⅡ_A-NLC在72 h内的累积释放量并绘制释放曲线,MTT法考察TⅡ_A-NLC对HaCaT细胞增殖的影响。结果 TⅡ_A-NLC的平均粒径为(178±9)nm,多分散度指数(PDI)为0.183±0.017,Zeta电位(-27.5±5.6)m V,体外72 h累积释放量为52.28%,TⅡ_A-NLC能显著降低TⅡ_A的光降解速率,TⅡ_A在一定范围内以剂量依赖性的方式抑制HaCaT细胞增殖,同质量浓度的TⅡ_A-NLC对HaCaT细胞增殖的抑制作用显著高于TⅡ_A溶液。结论 TⅡ_A-NLC稳定性好,具有良好的缓释作用,较好的细胞生物相容性,能显著提高TⅡ_A对HaCaT细胞增殖的抑制作用。
Objective To further evaluate the properties of tanshinone Ⅱ_A nanostructured lipid carrier (TⅡ_A-NLC) and investigate its effect on the proliferation of HaCaT cells. Methods The particle size and photostability of TⅡ_A-NLC were investigated by nanoparticle size analyzer and HPLC. The cumulative release of TⅡ_A-NLC in 72 h was determined by dialysis bag method and the release curve was drawn. MTT assay was used to examine the effect of TⅡ_A-NLC on HaCaT cells Effects of proliferation. Results The average diameter of TⅡ_A-NLC was (178 ± 9) nm, the PDI was 0.183 ± 0.017 and the Zeta potential was (-27.5 ± 5.6) mV. The cumulative release in vitro was 52.28% -NLC can significantly reduce the photodegradation rate of TⅡ_A. TⅡ_A inhibits the proliferation of HaCaT cells in a dose-dependent manner in a certain range. The inhibitory effect of TⅡ_A-NLC at the same concentration on the proliferation of HaCaT cells was significantly higher than that of TⅡ_A solution. Conclusion TⅡ_A-NLC has good stability, good sustained-release effect and good cell biocompatibility. It can significantly increase the inhibitory effect of TⅡ_A on the proliferation of HaCaT cells.
