本文探索研究了利用超薄层等电聚焦电泳进行燕麦品种鉴定所需的各种试验条件。结果显示:燕麦种子贮藏蛋白以醇溶蛋白为主,水溶蛋白次之,盐溶蛋白最少,因此应将醇溶蛋白作为燕麦品种鉴定的生化标记;一次性提取法与分步提取法相比,步骤较为简单,需时短,所得电泳谱带较多,更适用于燕麦种子蛋白提取;4种醇溶蛋白提取液中,以1%(V/V)巯基乙醇+1mol/L(W/V)尿素+乙二醇溶液所得电泳效果最佳,确定其为燕麦种子醇溶蛋白提取液以进一步研究。
This paper explores the various experimental conditions required for the identification of oat varieties using ultrathin layer isoelectric focusing electrophoresis. The results showed that: the main storage protein of oats was gliadin, followed by water-soluble protein and salt-soluble protein, so gliadin should be used as the biochemical marker for the identification of oat varieties. Compared with the step-by-step extraction method, The results showed that the electrophoresis bands were more suitable for the extraction of oat seed protein. The contents of 1% (v / v) mercaptoethanol + 1mol / L (W / V) The results of the best electrophoresis of urea + ethylene glycol solution, to determine the oat seed gliadin extract for further study.
