目的:比较巴戟天不同炮制品及提取部位的祛风湿作用,探讨其在细胞因子水平上的作用机制。方法:大鼠左后足跖皮内注射弗氏完全佐剂制备AA模型,分别灌胃给予巴戟天、巴戟肉、盐巴戟天、制巴戟天4种炮制品和巴戟天乙酸乙酯萃取部位、正丁醇萃取部位、剩余水部位和总多糖部位,以雷公藤多苷片为对照,在不同的时间点检测大鼠继发侧关节肿胀度;致炎28 d处死大鼠,采用ELISA法检测大鼠血清炎症细胞肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-2(IL-2)、干扰素-γ(IFN-γ)的水平。结果:与模型组比较,盐巴戟天及巴戟天乙酸乙酯部位、巴戟天正丁醇部位能明显减轻AA大鼠足肿胀度,降低异常升高的血清TNF-α、IL-1β、IL-6、INF-γ水平;而巴戟天、制巴戟天、巴戟天总多糖和巴戟天剩余水部位作用不明显。结论:盐巴戟天、巴戟天正丁醇部位和巴戟天乙酸乙酯部位对AA大鼠各项指标改善作用显著,效果优于巴戟天、制巴戟天、巴戟天总多糖和巴戟天剩余水部位,其作用机制可能与抑制促炎因子TNF-α、IL-1β、IL-6、INF-γ的产生有关。
OBJECTIVE: To compare the dispelling rheumatism effect of different processed products and parts of Morinda officinalis, and to explore its mechanism of action at cytokine level. METHODS: AA model was established by intradermal injection of Freund’s complete adjuvant in the left hind paw. Rats were administered with Morinda officinalis, Morinda officinalis, Morinda citrifolia, four kinds of processed products of Morinda officinalis and Morinda officinalis B Ester extraction, n-butanol extraction, residual water and total polysaccharides were used to detect the degree of secondary joint swelling at different time points; Serum levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), interleukin-6 (IL-6) and interleukin- 2), interferon-γ (IFN-γ) levels. Results: Compared with the model group, the ethyl acetate fraction of Morinda officinalis and Morinda officinalis, the n-butanol fraction of Morinda officinalis significantly reduced the degree of paw edema in AA rats, and decreased the levels of serum TNF-α, IL-1β, IL -6, INF-γ levels; while the Morinda officinalis, Morinda officinalis, Morinda officinalis and Morinda residual water role is not obvious. Conclusion: The effects of Morinda officinalis, Morinda officinalis, and ethyl acetate of Morinda officinalis on AA indexes are significant, and their effects are better than that of Morinda officinalis, Morinda officinalis, Morinda officinalis, The remaining water area of halberd could be related to the inhibition of the production of TNF-α, IL-1β, IL-6 and INF-γ.
