目的采用斑马鱼胚胎发育技术方法研究氯化两面针碱(Nitidine Chloride,NC)对斑马鱼胚胎的毒性效应。方法将4 hpf发育正常的胚胎分别置于一系列含有不同浓度NC的胚胎培养液中进行培养,测定不同发育时间点斑马鱼胚胎发育的畸形率、死亡率、LD50和ED50。结果随着药物浓度的增加,斑马鱼胚胎的死亡率、畸形率增加;随胚胎在含有药物的培养液中培养时间的增加,死亡率、畸形率增加。结论 NC的浓度与斑马鱼胚胎发育毒性存在明显的剂量-效应关系;NC作用时间长短与斑马鱼胚胎发育毒性亦存在明显的相关效应。
Objective To study the toxic effects of Nitidine Chloride (NC) on zebrafish embryo by zebrafish embryo development. Methods 4 hpf normal embryos were placed in a series of embryos culture medium containing different concentrations of NC for culturing. Deformity, mortality, LD50 and ED50 of zebrafish embryos at different developmental stages were determined. Results With the increase of drug concentration, the mortality and deformity rate of zebrafish embryos increased. The mortality and deformity rate increased with the increase of embryo culture time in drug-containing medium. Conclusion There is a significant dose-effect relationship between NC concentration and zebrafish embryonic developmental toxicity. There is also a significant correlation between the NC duration and zebrafish embryonic developmental toxicity.
