目的探讨妊娠期正己烷暴露对雄性仔代大鼠生殖发育的影响。方法 24只Wistar孕鼠随机分为4组,于孕期第1~20 d静式吸入0(对照组)、360、1 800、9 000 mg/m3正己烷;雄性仔鼠从出生后第9 d(PND9)开始称重,PND69处死,计算睾丸及附睾脏器系数;组织切片观察睾丸病理学改变;计算精子活率、活力、密度及畸形率;测定血清睾酮水平和睾丸丙二醛含量。结果正己烷9 000 mg/m3组雄性仔鼠体重增长明显低于对照组,睾丸脏器系数[(1.05±0.07)mg/g]低于对照组[(1.12±0.05)mg/g](P<0.05),精子活率和活力低于对照组;雄性仔鼠血清睾酮含量[(1.22±0.45)ng/m L]低于对照组[(2.53±0.99)ng/m L](P<0.05);正己烷9 000 mg/m3组雄性仔鼠睾丸组织出现曲精小管排列疏松、精原细胞萎缩、曲精小管管壁变薄等病理改变;各组雄性仔鼠睾丸丙二醛含量无明显差异。结论妊娠期高剂量正己烷暴露可通过非脂质过氧化途径介导明显抑制雄性仔代生殖发育。
Objective To investigate the effect of n-hexane exposure during pregnancy on the reproductive development of offspring rats. Methods Twenty-four pregnant Wistar rats were randomly divided into 4 groups: n = 360, 800, 000, 000 mg / m3 n-hexane. (PND9) were weighed and PND69 was sacrificed. The testicular and epididymal organ coefficients were calculated. Tissue sections were used to observe the pathological changes of testis. The sperm motility, viability, density and deformity rate were calculated. Serum testosterone levels and testis malondialdehyde contents were determined. Results The body weight of male offspring in n-hexane 9 000 mg / m3 group was significantly lower than that of control group (P <0.05), while the organ coefficient of testis was significantly lower than that of control group [(1.05 ± 0.07) mg / g <0.05), and the sperm motility and viability were lower than those of the control group. The serum testosterone level in male offspring was significantly lower than that of the control group [(1.22 ± 0.45) ng / m L [(2.53 ± 0.99) ng / m L] ). The testicular tissue of male offspring in n-hexane 9 000 mg / m3 showed loosely arranged tubules, atrophic spermatogonia and thinning of seminiferous tubules. The content of malondialdehyde difference. Conclusion Exposure to high-dose n-hexane during pregnancy can significantly inhibit the reproductive development of male offspring through the non-lipid peroxidation pathway.
