目的探讨针刺对急性脑缺血大鼠血液及缺血脑区炎性因子的影响。方法三月龄SD雌性大鼠100只,随机均分为正常组、假手术组、穴位组、非穴位组、模型组。采用开颅法电凝大脑中动脉复制急性脑缺血模型,针刺选用百会和人中穴,实验结束后采血、取脑组织,TTC染色测定梗死面积,放免法检测血液及缺血脑组织IL-1β、IL-6和IL-8含量。结果与正常组比较,模型组大鼠脑梗死范围明显,血液及缺血脑组织IL-1β、IL-6和IL-8含量显著升高(P<0.01);与模型组相比,穴位组大鼠梗死区域变小,血液及缺血脑组织IL-1β、IL-6和IL-8含量降低(P<0.05),但假电针组与模型组比较差异无统计学意义。结论针刺可以显著降低血液及缺血脑组织IL-1β、IL-6和IL-8含量,从而减轻其诱导的各种炎性细胞因子的表达,发挥抗脑缺血损伤作用。
Objective To investigate the effects of acupuncture on inflammatory factors in blood and ischemic brain regions of rats with acute cerebral ischemia. Methods 100 female SD rats of three months old were randomly divided into normal group, sham operation group, acupoint group, non-acupoint group and model group. The model of acute cerebral ischemia was created by electrocuring the middle cerebral artery by craniotomy. The rats were sacrificed by acupuncture at Baihui and Renzhong points. After the experiment, blood samples were collected and the infarct area was determined by TTC staining. The blood and brain tissue were detected by radioimmunoassay IL-1β, IL-6 and IL-8 levels. Results Compared with the normal group, the infarct size in the model group was significantly increased, and the levels of IL-1β, IL-6 and IL-8 in the blood and ischemic brain tissue were significantly increased (P <0.01). Compared with the model group, The area of infarction in rats decreased, and the levels of IL-1β, IL-6 and IL-8 in blood and ischemic brain tissue decreased (P <0.05), but there was no significant difference between the sham acupuncture group and the model group. Conclusion Acupuncture can significantly reduce the content of IL-1β, IL-6 and IL-8 in blood and ischemic brain tissue, so as to reduce the expression of various inflammatory cytokines induced by it and exert anti-ischemic effects.
