目的探讨顺铂(DDP)对宫颈癌淋巴转移作用的影响及其作用机制。方法将14只裸鼠建立人宫颈癌裸鼠移植瘤模型后,随机分为对照组和顺铂组各7只,对照组裸鼠腹腔注射PBS缓冲液,顺铂组裸鼠腹腔注射顺铂。观察裸鼠淋巴转移情况,采用病理学方法认证淋巴结及淋巴结内部的宫颈癌浸润,采用免疫组化方法检测肾小球足突细胞膜黏蛋白(Podoplanin)和胸苷磷酸化酶(thymidine phosphorylase,TP)的表达。结果顺铂组对淋巴转移有明显抑制作用,顺铂组淋巴转移个数及淋巴转移率均低于对照组(P<0.05)。顺铂组Podoplanin和TP的表达低于对照组(P<0.05)。结论应用顺铂治疗后,对裸鼠淋巴转移有明显的抑制作用。淋巴结中Podoplanin和TP的表达下调,抑制宫颈癌移植瘤的增殖和侵袭能力,可能是顺铂抑制宫颈癌淋巴转移的机制之一。
Objective To investigate the effect of cisplatin (DDP) on lymphatic metastasis of cervical carcinoma and its mechanism. Methods Twenty-four human nude mice bearing human cervical carcinoma were established and then randomly divided into 7 control and 7 cisplatin groups respectively. The control group was intraperitoneally injected with PBS buffer and the cisplatin group was intraperitoneally injected with cisplatin. The lymph node metastasis in nude mice was observed. The infiltration of cervical cancer in lymph nodes and lymph nodes was confirmed by pathology. Podoplanin and thymidine phosphorylase (TP) expressions in glomerular foot process were detected by immunohistochemistry. expression. Results The cisplatin group had a significant inhibitory effect on lymphatic metastasis. The numbers of lymphatic metastasis and lymphatic metastasis in cisplatin group were significantly lower than those in control group (P <0.05). The expression of Podoplanin and TP in cisplatin group was lower than that in control group (P <0.05). Conclusion After cisplatin treatment, lymphatic metastasis in nude mice was significantly inhibited. The down-regulation of Podoplanin and TP in lymph nodes may inhibit the proliferation and invasion of cervical cancer xenografts and may be one of the mechanisms by which cisplatin inhibits lymphatic metastasis of cervical carcinoma.
