目的:探讨8-溴-7-甲氧基白杨素(BrMC)诱导人非小细胞肺癌A549细胞凋亡作用及分子机制。方法:流式细胞术(PI染色)分别检测白杨素及BrMC处理后的细胞凋亡。蛋白印迹法检测BrMC对Bim蛋白表达以及N-乙酰半胱氨酸对BrMC介导Bim表达的影响。siRNA沉默Bim基因后流式细胞仪检测BrMC对细胞凋亡影响。结果:BrMC对A549细胞的凋亡作用较白杨素更强。经BrMC(10μmol/L)处理后,Bim蛋白水平呈时间依赖性增加。siRNA干扰后,BrMC诱导的A549细胞凋亡率明显降低。结论:BrMC通过上调Bim诱导肺癌A549细胞的凋亡。
AIM: To investigate the molecular mechanism of apoptosis induced by 8-bromo-7-methoxychitosan (BrMC) in human non-small cell lung cancer A549 cells. Methods: Flow cytometry (PI staining) were used to detect the apoptosis of chrysin and BrMC respectively. The effect of BrMC on Bim protein expression and N-acetylcysteine on BMC expression induced by BrMC was detected by Western blotting. Flow cytometry was used to detect the effect of BrMC on apoptosis after Bim gene silencing by siRNA. Results: The apoptosis of A549 cells induced by BrMC was stronger than that of chrysin. After treated with BrMC (10μmol / L), Bim protein level increased in a time-dependent manner. siRNA interference, BrMC induced A549 apoptosis rate was significantly reduced. Conclusion: BrMC can induce apoptosis of lung cancer A549 cells by up-regulating Bim.
