本研究以核素99mTc标记抗癌胚抗原 (CEA)单抗C5 0对肺癌模型的放射免疫导向手术 (RIGS)进行初步的探讨。根据有无肺转移 ,40只荷肺腺癌小鼠被随机配对分成两组 ,并经尾静脉注入 2 0 0 μCi/ 40 μg99mTc C5 0。注药后进行体外放免显像、手持型γ探测仪 (GDP)体内探测及井型探测仪分析。结果显示注药后 6h ,肿瘤显像最清晰。井型探测仪探测的肾脏与肿瘤的 %ID/ g高于其它脏器。GDP探测的肿瘤组织与正常组织的放射性比值 (T/NT)范围为 2 .2 39.73。肺转移癌的T/NT值为 2 .0 9± 0 .31。实验表明 ,99mTc C5 0对肺癌原发病灶及转移灶有较高的特异性和亲和力 ,利用GDP可准确区分肿瘤及正常组织并探测出微小转移病灶。
In this study, Radionuclide-guided surgery (RIGS) of lung cancer model was preliminary studied with radionuclide 99mTc labeled anti-CEA McAb C5 0. Forty lung adenocarcinoma-bearing lung adenocarcinoma mice were randomized into two groups according to the presence or absence of lung metastases and injected with 200 μCi / 40 μg of 99mTc C5 0 through the tail vein. In vitro radiopharmaceutical imaging, hand-held gamma detector (GDP) in vivo detection and well-type detector analysis. The results showed that 6h after injection, the most clear tumor imaging. Well-type detector to detect the kidney and tumor% ID / g higher than other organs. The ratio of radioactivity (T / NT) between tumor and normal tissue detected by GDP ranged from 2.39 to 3.73. The T / NT value of lung metastases was 2.09 ± 0.31. Experiments show that, 99mTc C5 0 of lung cancer primary lesions and metastases have a high specificity and affinity, the use of GDP can accurately distinguish the tumor and normal tissue and detect micrometastases.
