目的建立铵离子浓度的测定方法并应用于脂质体制备研究。方法将硫酸铵溶液加入显色剂显色后在630nm处比色测定,并考察了线性、精密度、稳定性、回收率等项目,测定脂质体透析液中的铵离子浓度。结果铵离子在0.95~9.50μg/mL范围内呈现良好的线性关系;回收率为101.29%,精密度与稳定性良好。通过测定铵离子确定了透析工艺。复合磷脂脂质体中的铵离子含量明显高于单一磷脂脂质体,与其载药后包封率的提高具有相关性。结论该方法适于空白脂质体透析液中铵离子含量的测定;复合磷脂脂质体提高马钱子碱载药量的机制与提高脂质体中铵离子的含量相关。
Objective To establish a method for the determination of ammonium ion concentration and apply it to the preparation of liposomes. Methods Ammonium sulphate solution was added to the color reagent and colorimetrically determined at 630 nm. The linearity, precision, stability, recovery and other items were examined to determine the ammonium ion concentration in the liposomal dialysate. Results The ammonium ion showed a good linear relationship in the range of 0.95-9.50 μg/mL; the recovery rate was 101.29%, and the precision and stability were good. The dialysis process was determined by measuring ammonium ions. The ammonium ion content in the composite phospholipid liposomes was significantly higher than that of the single phospholipid liposomes, which was related to the improvement of the entrapment efficiency after drug loading. Conclusion The method is suitable for the determination of ammonium ion content in dialysate of blank liposomes. The mechanism of enhancing the drug loading of strychnine by the composite phospholipid liposomes is related to the increase of ammonium ion content in the liposome.
