~(99m)Tc标记尿激酶(血栓显像剂),国内未见报道,我们于1984年研制成功,并应用于临床,得到较为满意效果。但尿激酶经过放射性核素标记以后是否仍然保持一定的生物活力,必须经过严格的活力测定才能判断。本文报道两种尿激酶活力测定方法:(1)气泡法,它是用时间差来进行计算的,即气泡上升时间减去放入水浴时间,将测试结果查标准曲线即得标记以后尿激酶效价单位。(2)纤维蛋白平板测定法,溶圈大小与尿激酶活力呈对数直线关系,在标准曲线上查出~(99m)Tc标记尿激酶效价单位。
~ (99m) Tc labeled urokinase (thrombus imaging agent), no domestic reports, we successfully developed in 1984 and applied to clinical, get more satisfactory results. But urokinase after radionuclide labeling still maintain a certain degree of biological activity, must undergo a rigorous determination of vitality to judge. This article reports two methods for measuring the activity of urokinase: (1) bubble method, which is calculated with the time difference, that is, the bubble rise time minus the time into the water bath, the test results check the standard curve was labeled after the urokinase titers unit. (2) fibrin plate assay, the size of the lysing circle and urokinase activity showed a logarithmically linear relationship, ~ (99m) Tc-labeled urokinase titer unit was found on the standard curve.
