目的:研究天花粉蛋白(TCS>)体内抗结肠癌作用。方法:将人结肠癌SW-1116移植于Swiss-Df品系8周龄裸小鼠肾包膜下,次日以不同剂量TCS注射入探小鼠腹膜腔内,以丝裂霉素(MMC)为阳性对照,生理盐水(NS)为阴性对照,每日给药一次,连续五日,观察天花粉蛋白对移植瘤体内抑制情况。结果:TCS剂量为0.75mg/kg组对移植瘤SW-1116的抑癌率为41.2%(P<0.001),较1.0mg/kgMMC组(抑癌率31.28%)为佳;0.5mg/kgTCS组抑癌率为27.50%(P<0.05),远小于TCS对小鼠的半数致死量(13.4mg/kg)。结论:TCS对结肠癌SW-1116裸鼠有体内抑癌作用。
Objective: To study the anti-colon cancer effect of trichosanthin (TCS>) in vivo. METHODS: Human colon cancer SW-1116 was transplanted into the kidney capsule of 8-week-old nude mice of Swiss-Df strain. The following day, different doses of TCS were injected into the peritoneal cavity of the mice. The mitomycin C was used as the MMC. The positive control and normal saline (NS) were negative controls. Once administered once a day for five consecutive days, the inhibitory effect of trichosanthin on tumor xenografts was observed. RESULTS: The tumor suppressor rate of transplanted tumor SW-1116 was 41.2% (P<0.001), which was better than 1.0 mg/kg MMC group (31.28% tumor suppressor rate). The tumor suppressing rate in kgTCS group was 27.50% (P<0.05), which was far less than the median lethal dose of TCS in mice (13.4 mg/kg). Conclusion: TCS has antitumor effect on colon cancer SW-1116 nude mice.
