目的:探讨丹皮酚(Paeonol)单用及联合5-氟尿嘧啶(5-FU)对人肝癌细胞系SMMC-7721的增殖抑制作用。方法:采用MTT法及光镜观察不同浓度Paeonol、5-FU单独和联合应用不同时间对SMMC-7721细胞增殖抑制作用,利用中效原理判定两药合用的效果。结果:两种药物单独应用时对SMMC-7721细胞的抑制作用呈明显的剂量依赖和时间依赖效应关系。两药合用大剂量时(大于中效浓度)为拮抗作用,小剂量(小于中效浓度)时为协同作用;两药给药时间及给药顺序影响效应大小。结论:Paeonol与5-FU均可抑制SMMC-7721细胞的增殖,两药合用大剂量时为拮抗效应,小剂量时为协同效应;给药时间及给药顺序可影响效应大小。
Objective: To investigate the inhibitory effects of paeonol and 5-fluorouracil (5-FU) on the proliferation of human hepatoma cell line SMMC-7721. Methods: MTT assay and light microscopy were used to observe the inhibitory effects of different concentrations of Paeonol and 5-FU on the proliferation of SMMC-7721 cells separately and in combination at different times. Results: The inhibition of SMMC-7721 cells alone or in combination with two drugs showed a dose-dependent and time-dependent effect. The combination of two large doses (greater than the concentration of the effect) for the antagonistic effect, a small dose (less than the concentration) is a synergistic effect; two drug administration time and administration order affect the size of the effect. CONCLUSION: Both Paeonol and 5-FU can inhibit the proliferation of SMMC-7721 cells. The combination of Paeonol and 5-FU can antagonize the effect of SMMC-7721 cells and the synergistic effect of SMMC-7721 cells.
