目的:观察活血熄风方对局灶性脑缺血模型大鼠神经细胞凋亡及Bcl-2、Bax蛋白表达的影响。方法:Wistar雄性大鼠40只随机分为5组:假手术组、模型组、活血熄风方治疗组(剂量分别为20,10,5 g.kg-1)。用光化学法建立局灶性脑缺血大鼠模型,造模前ig给药7 d,造模后24 h取材,用TUNEL法检测神经细胞凋亡数目,免疫荧光与激光共聚焦技术检测Bcl-2、Bax蛋白表达。结果:与模型组比较,活血熄风方各剂量组TUNEL阳性细胞数明显减少,Bcl-2蛋白表达升高,Bax蛋白表达降低,Bcl-2/Bax比值增高(P<0.01或P<0.05)。结论:活血熄风方具有明显的抗凋亡作用,其作用机制可能与增加Bcl-2蛋白表达,降低Bax蛋白表达,提高Bcl-2/Bax比值有关。
Objective: To observe the effect of Huoxue Xifeng Formula on neuronal apoptosis and expression of Bcl-2 and Bax proteins in rats with focal cerebral ischemia. METHODS: Forty Wistar male rats were randomly divided into 5 groups: sham operation group, model group, and Huoxue Xifeng Recipe treatment group (the doses were 20, 10, and 5 g.kg-1, respectively). A rat model of focal cerebral ischemia was established by photochemical method. The mice were given ig for 7 days before modeling and 24 hours after modeling. The number of apoptotic neurons was detected by TUNEL method. Bcl- was detected by immunofluorescence and laser confocal technique. 2. Bax protein expression. RESULTS: Compared with the model group, the number of TUNEL-positive cells in each dose group of Huoxue Xifeng Decoction significantly decreased, the expression of Bcl-2 protein increased, the expression of Bax protein decreased, and the Bcl-2/Bax ratio increased (P<0.01 or P<0.05). . Conclusion: Huoxue Xifeng Formula has obvious anti-apoptosis effect. Its mechanism may be related to increase of Bcl-2 protein expression, decrease of Bax protein expression and increase of Bcl-2/Bax ratio.
