目的:探讨小檗碱对高糖诱导的新生大鼠心肌成纤维细胞增殖及Akt激活影响。方法:分离新生乳鼠心肌成纤维细胞,MTT法检测不同浓度小檗碱(10~40μM)对高糖(25 mM)诱导心肌成纤维细胞增殖的影响。采用qPCR法检测大鼠心肌成纤维细胞纤连蛋白(FN)的表达。采用Western blot法检测大鼠心肌成纤维细胞中增殖细胞核抗原(PCNA)、Akt的表达。结果:25 mM高糖刺激48 h后心肌成纤维细胞的细胞增殖显著增加,FN、PCNA、p-Akt表达升高,小檗碱以剂量依赖性方式抑制高糖诱导上述指标的改变。结论:小檗碱可以通过抑制高糖诱导的心肌成纤维细胞增殖及胶原合成,其机制可能与抑制Akt有关。
Objective: To investigate the effects of berberine on the proliferation and Akt activation of neonatal rat cardiac fibroblasts induced by high glucose. Methods: The neonatal rat cardiac fibroblasts were isolated and the effects of different concentrations of berberine (10-40 μM) on the proliferation of cardiac fibroblasts induced by high glucose (25 mM) were determined by MTT assay. The expression of fibronectin (FN) in rat cardiac fibroblasts was detected by qPCR. Western blot was used to detect the expression of proliferating cell nuclear antigen (PCNA) and Akt in rat cardiac fibroblasts. Results: The proliferation of cardiac fibroblasts was significantly increased and the expression of FN, PCNA and p-Akt were increased after 48-M high glucose stimulation for 48 h. Berberine inhibited the changes of these indexes induced by high glucose in a dose-dependent manner. Conclusion: Berberine can inhibit the proliferation and collagen synthesis of cardiac fibroblasts induced by high glucose, and its mechanism may be related to the inhibition of Akt.
