目的:研究黄连解毒汤(HLJDT)全方及其各部位群抗炎活性,讨论HLJDT抗炎活性的物质基础。方法:通过鼠毛细管通透性和二甲苯致炎实验,建立动物模型以及确定黄连解毒汤的有效剂量,并依据全方的有效剂量计算出各组分群的给药剂量。将动物随机分为空白组,模型组,地塞米松组,HLJDT全方组,HLJDT上清液50%醇,HLJDT沉淀水洗,沉淀50%醇洗组,按传统给药方式连续3 d ig给药。末次给药60 min后尾静脉注射伊文思蓝(EB),然后立即进行小鼠二甲苯致炎实验,结合高效液相色谱(HPLC)研究HLJDT全方及其各部位群抗炎作用,比较各组间差异。结果:地塞米松,HLJDT全方,上清液50%醇洗和沉淀50%醇洗均能有效降低模型组的肿胀度,与模型组比较有显著性差异(P<0.01)。HLJDT全方,上清液50%醇洗和沉淀50%醇洗部位群均能有效降低耳片中EB含量,抑制肿胀度,EB含量分别为(11.8±1.8),(23.4±3.8),(25.2±3.2)ng·g-1;肿胀度抑制率分别为26.44%,29.89%,19.54%;HLJDT上清液50%醇洗抗炎作用比沉淀50%醇洗作用显著。结论:HLJDT,上清液50%醇洗和沉淀50%醇洗均能有效降低模型组的肿胀度,结合HPLC结果可推断,黄酮与生物碱相互作用后,可以降低生物碱副作用,增加环烯醚萜类成分后,抗炎效果更显著,为抗炎有效部位。
OBJECTIVE: To study the anti-inflammatory activity of HLJDT and its parts, and to discuss the material basis of anti-inflammatory activity of HLJDT. Methods: The mouse model of capillary permeability and xylene toxicity was established. The animal model was established and the effective dosage of Huanglian Jiedu Decoction was determined. The dosage of each group was calculated according to the effective dose of the compound. The animals were randomly divided into blank group, model group, dexamethasone group, HLJDT whole group, HLJDT supernatant 50% alcohol, HLJDT precipitation washing, sedimentation 50% alcohol wash group, according to the traditional way of administration for 3 d medicine. Evans Blue (EB) was injected through the caudal vein 60 min after the last administration, and then the mouse xylene-induced inflammatory reaction was performed immediately. The anti-inflammatory effects of HLJDT and its fractions were studied by high performance liquid chromatography (HPLC) Differences between groups. Results: Dexamethasone, HLJDT, 50% alcoholic wash and 50% alcohol wash of HLJDT could effectively reduce the degree of swelling of the model group, which was significantly different from the model group (P <0.01). HLJDT, 50% alcohol supernatant and 50% ethanol precipitation could effectively reduce the content of EB and inhibit the degree of swelling. The content of EB was (11.8 ± 1.8), (23.4 ± 3.8), ( 25.2 ± 3.2) ng · g-1. The inhibition rates of swelling degree were 26.44%, 29.89% and 19.54% respectively. The anti-inflammatory effect of 50% alcohol solution of HLJDT supernatant was more significant than that of 50% ethanol precipitation. CONCLUSION: HLJDT, 50% alcoholic wash and 50% ethanol wash can effectively reduce the degree of swelling in model group. Combined with HPLC results, it can be concluded that the interaction between flavonoids and alkaloids can reduce the side effects of alkaloids, After the ether terpenoids, the anti-inflammatory effect is more significant, an effective anti-inflammatory sites.
