目的:开发利用新疆细虫草资源,为新疆细虫草相关真菌的研究和开发利用提供基础资料。方法:研究从新疆细虫草菌核中分离得到一种白色菌株,编号为SFYT002,采用形态学观察和ITS以及18SrDNA测序,在GenBank比对,进行系统进化树分析,并采用滤纸片法研究其体外抗菌作用。结果:菌株SFYT002主要的特征为菌落白色棉绒状,瓶梗长在分枝的分生孢子梗上,瓶梗基部膨大,顶部急剧变细,部分颈部弯曲,孢子小且透亮,孢子有出芽现象,常形成聚集孢子头,分生孢子球形,亚球形至椭圆形。ITS和18SrDNA序列分析表明,弯颈霉属为单系发生,菌株SFYT002与膨大弯颈霉聚为一支。研究其抑菌作用表明,菌株SFYT002发酵液及其不同极性的提取物对4种常见致病菌,均有很强的抑制作用。结论:通过形态学和分子生物学特征将菌株SFYT002鉴定为膨大弯颈霉,具有较强的抑菌活性。
OBJECTIVE: To develop and utilize the Xinjiang Cordyceps resources and provide the basic information for the research, development and utilization of the related fungi of Cordyceps sinensis in Xinjiang. Methods: A white strain isolated from Xinjiang Cordyceps nucleus was isolated and identified as SFYT002. Phylogenetic tree, ITS and 18S rDNA sequencing were used to analyze the phylogenetic tree in GenBank. Antibacterial effect. Results: The main feature of strain SFYT002 was white lint of colonies. The bottle stem was long on the branchy conidiophore. The base of the bottle stem expanded, the top of the bottle became sharply thinned, the part of the neck was bent, the spores were small and translucent, and the spores were sprouting Phenomenon, often formed aggregated spore head, conidia spherical, sub-spherical to oval. ITS and 18S rDNA sequence analysis showed that the genus Pholiota was single-line genus, and strain SFYT002 was aggregated with Penicillium inconginius. The study of its bacteriostasis showed that the strain SFYT002 fermentation broth and its extracts of different polarity had a strong inhibitory effect on four kinds of common pathogens. Conclusion: The strain SFYT002 is characterized by morphological and molecular biological features and is characterized by its strong antibacterial activity.
