目的:研究桂郁金水提物对大鼠肝胞浆液和微粒体内Ⅱ脱毒酶的影响。方法:雄性SD大鼠,桂郁金水提液按(生药剂量)0.81,2.43,7.29 g·kg-13个剂量组,灌胃给药,1次/d,连续3周,末次给药后,动物取血,分离血清,测丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、尿素氮(BUN)、肌酐(Cr),摘除肝脏,差速离心法制备肝脏胞浆液及微粒体,测定胞浆液的过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽-过氧化物酶(GSH-Px)、谷胱甘肽还原酶(GR)和微粒体内谷胱甘肽-S-转移酶(GST)和尿苷二磷酸-葡萄糖醛酸转移酶(UGT)的活性。结果:桂郁金水提液对大鼠体重增长和肝脏指数及血清ALT,AST含量均无明显影响;桂郁金各剂量组明显降低Cr含量(P<0.01);中、高剂量组明显降低了BUN含量(P<0.01)。在肝胞浆液,与对照组比较,桂郁金各剂量组对CAT和GR的活性没有明显影响;桂郁金各剂量均明显增高GSH-Px活性(P<0.01);桂郁金中、高剂量明显增高SOD活性(P<0.01,P<0.05)。在肝微粒体中,与对照组比较,桂郁金各剂量组对UGT的活性没有显著的影响;桂郁金各剂量均明显增高GST活性(P<0.05)。结论:桂郁金对大鼠肝脏和肾脏没有毒性,对肾脏可能还具有保护作用。桂郁金可诱导胞浆液和微粒体内脱毒酶的功能,具有抗氧化作用,并能加速体内毒性代谢物的排除,增加肝脏的解毒能力。
OBJECTIVE: To study the effects of aqueous extract of Guanyujin on Ⅱ cytosolic detoxification enzyme in rat liver cytosol and microsome. METHODS: Male Sprague Dawley rats and Guiyujin aqueous extract were intragastrically administered once a day for 3 weeks at doses of (crude drug) 0.81,2.43,7.29 g · kg-13. After the last administration, Blood was taken from the animals, serum was separated, ALT, BUN and Cr were measured, liver was removed, and liver cytosol and microparticles were prepared by differential centrifugation The activities of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), glutathione reductase (GR) In vivo glutathione-S-transferase (GST) and uridine diphosphate-glucuronosyltransferase (UGT) activity. Results: The aqueous extract of Guiyujin had no significant effect on body weight gain, liver index and serum ALT, AST content in the rats; the content of Cr in the Guiyujin groups significantly decreased (P <0.01); the medium and high dose groups significantly decreased BUN content (P <0.01). In the liver cytoplasm, compared with the control group, Guanyujin each dose group had no significant effect on the activity of CAT and GR; each dose of Guiyu gold significantly increased GSH-Px activity (P <0.01); Guiyu gold medium and high dose Significantly increased the activity of SOD (P <0.01, P <0.05). In liver microsomes, compared with the control group, each dose of Guiyu gold had no significant effect on the activity of UGT; each dose of Guiyu gold significantly increased the activity of GST (P <0.05). Conclusion: Guanyujin is not toxic to the liver and kidney of rats, and may also have a protective effect on the kidneys. Guiyu can induce cytosolic fluid and microsomal detoxification enzyme function, with anti-oxidation, and can speed up the exclusion of toxic metabolites in the body, increase liver detoxification.
