马铃薯四倍体(2n=4x=48)栽培种花药培养,在MS培养基的基础上,添加激动素0.5毫克/升、2,4~D1毫克/升。花药于花粉细胞处于单核靠边期接种,诱导出愈伤组织和胚状体。培养基中添加IAA0.5毫克/升和6-BA1毫克/升有利于分化成苗,再添加NAA0.05毫克/升,生根素0.2毫升/升,则对幼苗生根和健壮有较好效果。花药培养以胚状体诱导成苗比愈伤组织诱导成苗的高。通过细胞学观察,愈伤组织分化成苗,有混倍体(2n=28、2n=32)、单倍体( 2n=24)和四倍体( 2n=48),而从胚状体诱导成苗均为双单倍体( 2n=24)。另外,对花药培养在马铃薯育种和实生种子利用方面进行了研究和讨论。
Potato tetraploid (2n = 4x = 48) cultivars anther culture, on the basis of MS medium, add kinetin 0.5 mg / l, 2,4 ~ D1 mg / l.
