目的:为研究肿瘤坏死因子α(TNF-α)相关的脑缺血再灌注损伤机制和相关药物提供参考。方法:以“脑缺血”“细胞凋亡”“炎症反应”“TNF-α”“Apoptosis”“Inflammation”“Cerebral ischemia”等为关键词,组合查询2004-2015年在Pub Med、中国知网、万方等数据库中的相关文献,对TNF-α在脑缺血再灌注损伤中的机制及相关药物进行综述。结果与结论:共检索到相关文献130余篇,其中有效文献43篇。TNF-α几乎参与了脑缺血再灌注损伤的全过程,是脑缺血再灌注损伤的重要原因之一,可以导致脑水肿、血脑屏障损伤、炎症反应、细胞凋亡等。目前,TNF-α相关的抗脑缺血药物研究主要有抗炎药物、抗细胞凋亡药物以及作用广泛的各类植物提取物等。很多药物研究揭示了其抗脑缺血过程中降低缺血区TNF-α水平的效应,并通过此效应降低脑缺血再灌注损伤,发挥了抗脑缺血作用。TNF-α相关的脑缺血再灌注损伤机制研究和药物研究已逐步展开,而TNF-α的双向作用有待进一步研究。
Objective: To provide a reference for studying the mechanism of cerebral ischemia-reperfusion injury associated with tumor necrosis factor α (TNF-α) and related drugs. METHODS: The key words were “cerebral ischemia”, “apoptosis”, “inflammatory response” “TNF-α”, “Apoptosis”, “Inflammation” and “Cerebral ischemia” , A combination of 2004-2015 in the Pub Med, China National Knowledge Network, Wanfang database and other relevant literature on TNF-α in cerebral ischemia-reperfusion injury mechanisms and related drugs are reviewed. RESULTS AND CONCLUSION: More than 130 articles have been retrieved, of which 43 are available. TNF-alpha is involved in almost the whole process of cerebral ischemia-reperfusion injury and is one of the important causes of cerebral ischemia-reperfusion injury. It can cause cerebral edema, blood-brain barrier injury, inflammatory reaction and apoptosis. Currently, there are anti-inflammatory drugs, anti-apoptotic drugs and various kinds of plant extracts with a wide range of effects on TNF-α-related anti-cerebral ischemia drugs. Many drug studies have revealed its anti-ischemic ischemic area of ?? reducing the level of TNF-a effect, and through this effect reduces cerebral ischemia-reperfusion injury, play a role in anti-cerebral ischemia. TNF-α-related mechanisms of cerebral ischemia-reperfusion injury and drug research has gradually started, and the two-way role of TNF-α remains to be further studied.
