[目的]研究仁术健脾理气方对碘乙酰胺(iodoacetamide,IA)诱导的功能性消化不良(FD)大鼠胃排空功能及胃促生长素(Ghrelin)、5-羟色胺(5-HT)、降钙素基因相关肽(CGRP)的影响,初步探讨仁术健脾理气方治疗FD的机制。[方法]雄性10日龄SD幼鼠100只,随机分为正常组(30只)和造模组(70只)。采用0.1%IA灌胃诱导大鼠FD模型。大鼠8周龄时进行模型评定,将造模成功的70只大鼠随机分为模型组(30只)、中药低剂量组(10只)、中药中剂量组(10只)、中药高剂量组(10只)和阳性药组(10只)5组。正常组与模型组予以0.9%氯化钠溶液1ml/100g灌胃,中药低、中、高剂量组分别给予生药含量0.477 5g/ml、0.955 5g/ml、1.911g/ml的中药煎剂1ml/100g灌胃,阳性药组给予0.3mg/ml多潘立酮溶液1ml/100g灌胃。采用苏木精-伊红染色观察大鼠胃组织病理变化。采用放射免疫法检测大鼠胃组织髓过氧化物酶(MPO)活性以及血清中Ghrelin、5-HT、CGRP的含量。[结果]胃排空结果显示,中药高剂量组和阳性药组大鼠3h摄食量和胃排空率显著高于模型组(P<0.05),而3h后胃内食物残余量显著低于模型组(P<0.05)。血清学结果显示,中药各剂量组和阳性药组FD大鼠血清中Ghrelin和5-HT的含量均不同程度地升高(均P<0.05),且高剂量组与阳性药组比较差异无统计学意义;中药各剂量组和阳性药组FD大鼠血清中CGRP的含量均不同程度地降低(均P<0.05),且高剂量组与阳性药组比较差异无统计学意义。[结论]仁术健脾理气方对IA诱导的FD大鼠具有加快胃排空、降低胃肠感觉过敏的作用,其机制可能与外周血Ghrelin、5-HT含量升高、CGRP含量降低有关。
[Objective] To investigate the effect of Renzhu Jianpi Liqi on gastric emptying in rats with functional dyspepsia (FD) induced by iodoacetamide (IA) and the effects of Ghrelin, 5-HT ) And calcitonin gene-related peptide (CGRP), and to explore the mechanism of Ren Shu Jianpi Liqi Decoction in the treatment of FD. [Methods] One hundred male 10-day-old SD rats were randomly divided into normal group (30 rats) and model group (70 rats). Induction of FD model in rats by intragastric administration of 0.1% IA. The rats were evaluated at 8 weeks of age. The 70 successful rats were randomly divided into a model group (30 rats), a low-dose Chinese medicine group (10 rats), a middle-dose Chinese herb group (10 rats) Group (10) and positive drug group (10) 5 groups. The normal group and the model group were given 0.9% sodium chloride solution 1ml / 100g gavage, the Chinese medicine low, medium and high dose groups were given crude drug content of 0.477 5g / ml, 0.955 5g / ml, 1.911g / ml of traditional Chinese medicine decoction 1ml / 100g gavage, positive drug group given 0.3mg / ml domperidone solution 1ml / 100g gavage. The pathological changes of gastric tissue were observed by hematoxylin-eosin staining. The activity of myeloperoxidase (MPO) and the contents of Ghrelin, 5-HT and CGRP in the stomach of rats were detected by radioimmunoassay. [Results] The results of gastric emptying showed that the 3 h intake and the gastric emptying rate of rats in the high-dose and high-dose groups were significantly higher than those in the model group (P <0.05), while the residual gastric food volume was significantly lower than the model group after 3 h Group (P <0.05). The results of serology showed that the content of Ghrelin and 5-HT in the serum of FD rats in each dose group and the positive drug group increased to some extent (all P <0.05), and there was no statistical difference between the high dose group and the positive drug group The content of CGRP in serum of FD rats in each dose group and the positive drug group decreased to some extent (all P <0.05), and there was no significant difference between the high dose group and the positive drug group. [Conclusion] Renzhan Jianpi Liqi Decoction can accelerate gastric emptying and reduce gastrointestinal hypersensitivity in IA induced FD rats. The mechanism may be related to the increase of Ghrelin and 5-HT content and the decrease of CGRP content in peripheral blood.
